Basically, Espada got Mike to promise that if he got 500 likes on a tweet using this Twitter Message as the image, then DeCarlo voice a line of Espada's choosing as Hugh Neutron.
The trailer does make it clear that all characters do have a signature weapon in some capacity. If these items are a part of the gameplay mechanics, it would be great if you could also customize them to some extent. In order to compete with genre juggernauts, Nickelodeon: All-Stars Brawl could easily set itself apart with weapons or items that are attached to specific characters.
And, this Tweet is making a joke about Jimmy Neutron's "alternate costumes" getting leaked as if this were real or the game was already out. But, frankly, we do hope that All-Stars Brawl has alternate costumes and colors, as it would be an easy way for the developers to tug at our nostalgia even more than they already have.
